重磅!张锋团队:1小时内检测新冠病毒

src="image/20201014/ed71549268b7ac686346d283d56fbd4b_2.png" /></p></p><p><br  /></p><p><span>SHERLOCK,由张锋开发并命名,同时,SHERLOCK是神探夏洛克·福尔摩斯(Sherlock Holmes)的名字。SHERLOCK技术由张锋团队于2017年发明,此后在拉沙热、埃博拉、寨卡病毒、登革热等RNA病毒疫情检测中大显身手。随后,SHERLOCKv2 诞生了,与 SHERLOCK 相比,其灵敏度提高了100倍,并能在同一样品中检测出多种病毒感染,比如能同时检出寨卡和登革热病毒。</span></p><p><span><br  /></span></p><p><p><img src="image/20201014/d76e350a52fc69aaa34f116719f66cae_3.jpg" /></p></p><p><span></span><br  /></p><p><span>之所以能达到这样效果,是因为他们在 SHERLOCKv2 中引入了多种来自不同种属细菌的 Cas13 酶,如 LwaCas13a 和 PsmCas13b。Cas13是一种依赖于RNA靶向的RNA核酸酶。世界上许多最常见或致命的人类病原体都是RNA病毒(例如埃博拉病毒、艾滋病病毒,流感病毒、SARS病毒等)此次在武汉爆发的新型冠状病毒,同样是RNA病毒。</span></p><p><span><br  /></span></p><p><span>针对此次新型冠状病毒,研究设计了两个向导RNA(gRNA),分别用于识别新冠病毒的S基因和Orf1ab基因。一旦检测样本中含有新型冠状病毒,向导RNA就会识别到它的S基因和Orf1ab基因,然后形成视觉可见的结果。</span></p><p><span><br  /></span></p><p><p><img
